Product Specific Information

The monoclonal antibody P1H12 recognizes CD146 (also known as MUC18, s-endo, Endo-CAM, and Mel-CAM), a member of the immunoglobulin superfamily. CD146 is expressed on endothelial cells, bone marrow fibroblasts, some tumors (especially melanoma), mesenchymal stromal cells, and endometrial stromal cells. It is also found on a subset of T cells and activated lymphocytes. The protein is heavily glycosylated, with more than 50% of its mass from carbohydrates.

This antibody cross-reacts with mouse, rabbit, and canine, but not rat.

Each product includes:

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated using normal human peripheral blood cells and HUVEC. Recommended usage: 5 µL (0.02 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Compatibility Notice:
NovaFluor dyes are incompatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI). Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.

CellBlox Plus Blocking Buffer:
Updated formulation (Cat. No. C001T06F01) for optimal staining. Add 5 µL per sample, regardless of the number of NovaFluor conjugates used. For single-color controls, use 5 µL per 100 µL cell sample (10³–10⁸ cells).

Target Information

CD146 (MCAM) is an integral membrane glycoprotein of the immunoglobulin superfamily, expressed at cell-cell junctions in endothelial cells and various normal and malignant cell types. It functions as a calcium-independent adhesion molecule and signal transduction mediator, recruiting Fyn kinase and promoting tyrosine phosphorylation of intracellular proteins involved in actin cytoskeleton assembly.

CD146 supports endothelial cohesion, contributes to melanoma cell vascular interactions and metastasis, and may play roles in neural crest cell adhesion during embryonic development. It acts as a surface receptor triggering phosphorylation of FYN and PTK2, leading to transient increases in intracellular calcium.
