Product Specific Information

The SIDI8BEE monoclonal antibody reacts with human and rhesus macaque CD8 beta. The CD8b chain associates with the CD8a chain to form the CD8ab heterodimer expressed on thymocytes and peripheral cytotoxic T cells. The CD8 receptor complex binds to MHC class I and plays a role in T cell development and activation.

SIDI8BEE detects CD8 beta chain in both CD8ab heterodimer and CD8bb homodimer. It is cross-reactive to human and rhesus macaque CD8b. Other non-human primates have not been tested.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells.
Use 5 µL (0.2 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., propidium iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining.
• Add 5 µL per sample to antibody cocktails before combining with cells.
• For single-color controls, use 5 µL per 100 µL of cell sample.

Target Information

CD8B is a cell surface glycoprotein found on cytotoxic T lymphocytes, functioning as a coreceptor with T-cell receptor for antigen recognition in the context of class I MHC molecules. The functional receptor may be a homodimer (alpha-alpha) or heterodimer (alpha-beta). The gene encodes CD8 beta chain isoforms, with multiple alternatively spliced variants described. A pseudogene is also located on chromosome 2.
