Target Information

Glycogen synthase kinase-3 (GSK3) is a protein kinase originally identified as a regulator of glycogen synthase, a key enzyme in glycogen metabolism. It is involved in regulation of various cellular functions including protein synthesis, cell proliferation, differentiation, microtubule dynamics, and apoptosis.

GSK3’s substrate specificity requires a pre-existing phosphoserine or phosphotyrosine four residues C-terminal to the phosphorylation site. Two isoforms exist — GSK3α and GSK3β — regulated by phosphorylation. Phosphorylation of GSK3β on Ser9 (Ser21 in GSK3α) by protein kinase B (PKB) inactivates the enzyme, mediating growth factor inhibition. Activation occurs via phosphorylation of Tyr216 (Tyr279 in GSK3α).

Activated GSK3β phosphorylates multiple cellular proteins including p53, c-Myc, c-Jun, HSF-1, and cyclin D1. It also phosphorylates tau protein at abnormal sites, contributing to Alzheimer’s disease progression.
