Product Specific Information
Description:
The 9E2 monoclonal antibody reacts with human CD335 (NKp46), a member of the natural cytotoxicity receptor family (NKp30, NKp44). CD335 is expressed on NK cells and functions as a type I transmembrane glycoprotein involved in activating and expanding resting NK cells through interaction with dendritic cells, contributing to the elimination of tumor and virus-infected cells. It also interacts with pancreatic beta cells during type 1 diabetes development. The 9E2 antibody has been reported to induce cytotoxicity.
Each product includes one vial of NovaFluor conjugate and one vial of CellBlox Plus Blocking Buffer.

Applications Tested:
- Pre-titrated and tested on normal human peripheral blood cells
- Recommended use: 5 µL (0.125 µg) per test (final volume 100 µL, 10⁵–10⁸ cells/test)
Important Notes:
- NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, and DAPI.
- Use Invitrogen LIVE/DEAD Fixable Dead Cell Stains for viability analysis with NovaFluor dyes.
- CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) must be used for optimal staining.
- Add 5 µL per sample before combining with cells.
- For single-color controls, use 5 µL per 100 µL of cell sample (10³–10⁸ cells).

Target Information
NKp46 (CD335, NCR1) is a cytotoxicity-activating receptor expressed on NK cells, enhancing their ability to mediate tumor cell lysis. It contains two Ig-like domains related to leukocyte immunoglobulin-like (LIR) and killer inhibitory receptors (KIR).
