Product Specific Information

Description:
The eBioRDR5 monoclonal antibody reacts with human CD127 (Interleukin-7 Receptor alpha). CD127 complexes with CD132 (common gamma chain, γc) to form the IL-7 receptor (IL-7R). CD127 is a type I glycoprotein (75–80 kDa) expressed by immature B cells, thymocytes, and most mature T cells, with transient down-regulation upon activation. IL-7 binding triggers signaling through tyrosine kinase pathways including Jak/STAT, essential for lymphocyte development and T-cell homeostasis.

CD127 expression is down-regulated on CD4⁺CD25⁺ regulatory T cells (Tregs). CD4⁺CD25⁺CD127^low cells are enriched for Foxp3⁺ Tregs with suppressive activity. Binding of eBioRDR5 to PBMCs is blocked by pre-incubation with recombinant human IL-7 (Product #14-1079-80).

Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested by flow cytometry of normal human peripheral blood cells. Use 5 µL (0.5 µg) per test (100 µL final volume; 10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Violet™ 421 (BV421) emits at 423 nm; use with a violet (405 nm) laser. Ensure cytometer compatibility.

When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (SB-4401-42) or Brilliant Stain Buffer™ (00-4409-75) to minimize non-specific interactions.

Target Information

CD127 (Interleukin-7 Receptor alpha, IL7Rα) is a glycoprotein regulating lymphopoiesis. The active IL7R receptor is an alpha/gamma chain heterodimer. The gamma(c) chain activates signal transduction, while the alpha chain mediates specific signaling events. CD127 promotes proliferation of precursor B cells, thymocytes, T cell progenitors, and mature CD4⁺/CD8⁺ T cells. Dysregulation is associated with severe combined immunodeficiency and related disorders.
