Product Specific Information

The 2B8 monoclonal antibody reacts with mouse CD117 (c-Kit receptor), also known as Steel factor receptor or stem cell factor receptor. This 145-kDa tyrosine kinase receptor is expressed by hematopoietic progenitor cells in mouse bone marrow and by mast cells. Interaction with steel factor promotes proliferation and differentiation of hematopoietic progenitor cells.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse bone marrow cells at ≤0.25 µg/test (100 µL final volume). Optimal titration recommended.

Brilliant Violet™ 421 (BV421): Emits at 423 nm; suitable for cytometers with 405 nm violet laser.
When using multiple polymer dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Target Information

KIT (c-KIT) is a proto-oncogene encoding a type 3 transmembrane receptor for MGF (mast cell growth factor, stem cell factor). KIT regulates growth and activation of hematopoietic and non-hematopoietic cells. Mutations in KIT are associated with gastrointestinal stromal tumors, mast cell disease, acute myelogenous leukemia, and piebaldism.
c-Kit is expressed in hematopoietic stem cells, germ cells, mast cells, and gastrointestinal tract cajal cells. Binding of SCF induces receptor dimerization and autophosphorylation, activating downstream pathways (Ras/ERK, PI3K, PLC-γ, JAK/STAT). KIT activation is essential for hematopoiesis, stem cell maintenance, and gametogenesis.
