Product Specific Information
The eBioH4A3 monoclonal antibody reacts with human CD107a (LAMP-1), a highly glycosylated 110 kDa protein primarily located in lysosomal/endosomal membranes. CD107a is transiently expressed on the surface of degranulating cytolytic T cells and upregulated on activated platelets and some cancer cells.
- Applications Reported: Intracellular staining followed by flow cytometric analysis
- Applications Tested: Pre-diluted and tested for intracellular staining of Jurkat cells using the Intracellular Fixation & Permeabilization Buffer Set (Product #88-8824-00)
- Recommended Use: 5 µL (0.5 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test)

Brilliant Violet™ 421 (BV421) emits at 423 nm and is suitable for cytometers equipped with a 405 nm violet laser.
Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies (e.g., Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Product #SB-4401-42) or Brilliant Stain Buffer™ (Product #00-4409-75) to reduce nonspecific interactions.

Target Information
LAMP1 (CD107a) is a major lysosomal membrane component, with 1–2% found on the plasma membrane. It is a heavily glycosylated type I integral membrane protein involved in lysosome biogenesis and fusion with phagosomes. Upon cell activation, LAMP1 translocates to the plasma membrane via a tyrosine-based motif (YXXI). Alterations in this motif affect lysosome localization.
Cell surface LAMP1 and LAMP2 promote PBMC adhesion to vascular endothelium, suggesting a role in inflammatory cell recruitment. Increased LAMP1 expression is observed in neurons and glial cells near senile plaques in Alzheimer’s disease and in thymic involution.
