Invitrogen™ LentiArray™ Human Cancer Biology CRISPR Library

The Invitrogen™ LentiArray™ Human Cancer Biology CRISPR Library targets 510 genes commonly involved in the development of cancer, with up to 4 gRNAs per gene (pooled in a single well) for a total of 2,040 gRNAs. Libraries are supplied as 100 μL of ready-to-use lentiviral particles per gene target, with an average titer of 1×10⁸ TU/mL (functional titer determined by antibiotic resistance).

LentiArray CRISPR libraries are designed using Thermo Fisher Scientific’s proprietary CRISPR gRNA design algorithm.
This algorithm ensures:

• High editing efficiency and specificity
• Knockout of all known isoforms of each target gene
• Up to four high-quality gRNAs per gene for robust knockout across diverse cell types

Experimental Flexibility

The LentiArray CRISPR system uses a two-vector design, expressing Cas9 nuclease and gRNA on separate lentiviral constructs.
This allows you to:

• Control timing and method of genome editing delivery
• Co-infect cells with both Cas9 and gRNA lentiviruses
• Establish stable Cas9-expressing cell lines

The Cas9 lentivirus expresses a human, codon-optimized Cas9 nuclease with a blasticidin-resistance gene under the EF-1α (EFS) promoter.
The gRNA lentiviruses include a puromycin resistance gene for enrichment of edited cells, enabling stronger phenotypes.

Fluorescent markers are not included, allowing compatibility with any reporter wavelength and enabling multiplexed experiments.
