Specificity Comments:
The ATG5 protein, in combination with autophagy protein 12, acts as an E1-like activating enzyme in a ubiquitin-like conjugating system. It plays roles in autophagic vesicle formation, mitochondrial quality control after oxidative damage, negative regulation of innate antiviral immune response, lymphocyte development, MHC II antigen presentation, adipocyte differentiation, and apoptosis. ATG5 is essential for autophagy, allowing cells to degrade their own components when no longer useful.

Target Information

ATG5 (Autophagy Related 5) is crucial for autophagy and may participate in apoptosis. It is involved in mitochondrial quality control, antiviral immune regulation, lymphocyte proliferation, MHC II antigen presentation, and adipocyte differentiation. Upon conjugation to ATG12, the complex aids autophagosome formation. ATG5 interacts with FADD to contribute to autophagic cell death. The ATG5-ATG12 conjugate forms an isolation membrane that detaches before or after autophagosome completion. APG5 expression occurs downstream of caspase activity and associates with innate immune proteins such as RIG-I and VISA, inhibiting type I interferon production and facilitating viral replication. Dysfunction of ATG5 is linked to spinocerebellar ataxia.
