Product Specific Information

The PC61.5 antibody reacts with mouse CD25, the 55 kDa interleukin-2 receptor alpha chain (IL-2Rα). CD25 is expressed by early progenitors of T and B lineage as well as by activated mature T and B lymphocytes. CD25 binds IL-2 with low affinity alone but forms a high-affinity receptor with CD122 (IL-2Rβ) and CD132 (common γ chain). Binding of IL-2 to both receptor classes is inhibited by the PC61.5 antibody. CD25 is involved in lymphocyte differentiation, activation, and proliferation.

• Applications Tested: Tested on mouse splenocytes at ≤0.5 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimal titration recommended.
• Fluorochrome: Brilliant Ultra Violet™ 805 (BUV805), emits at 797 nm, for UV (355 nm) laser-equipped cytometers.
• Light Sensitivity: Protect from light; sensitive to photo-induced oxidation.
• Fixation: Compatible with IC Fixation Buffer or 1-step Fix/Lyse Solution for up to 3 days at 4°C in dark. Short-term methanol fixation is possible (<1 hour).
• Recommended Buffers: Use Super Bright Complete Staining Buffer (SB-4401) or Brilliant Stain Buffer (00-4409-75) to minimize polymer dye interactions.

Target Information

CD25 (IL2RA) encodes the interleukin-2 receptor alpha chain, part of a heterotrimeric receptor complex with IL2RB and IL2RG. It mediates IL-2 signaling critical for T and B cell proliferation, NK cell activation, and immune regulation. Mutations in IL2RA are associated with immune dysregulation and interleukin-2 receptor alpha deficiency. In mice, gene disruption leads to colitis-like disease, indicating its role in immune homeostasis.
