The monoclonal antibody RDP4ND4 recognizes human interferon regulatory factor 7 (IRF7), a 54 kDa transcription factor essential for antiviral innate immune response and activation of type I IFN genes. IRF7 is constitutively expressed in plasmacytoid dendritic cells, B cells, and monocytes, and is strongly induced upon viral infection or interferon stimulation.
IRF7 activates IFN-β and IFN-α genes via virus-activated (MyD88-independent) or TLR-activated (MyD88-dependent) pathways. Upon virus exposure, IRF7 is induced through TLR7, TLR8, and TLR9, promoting IFN gene expression. Phosphorylation of IRF7 triggers dimerization and nuclear translocation, enabling activation of target genes. Expression is upregulated by interferon through the Jak-STAT pathway in a positive feedback loop.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-diluted and tested for flow cytometry using stimulated normal human peripheral blood cells with the Foxp3/Transcription Factor Staining Buffer Set (Product #00-5523-00) and protocol.
Recommended protocol: BestProtocols® – Protocol B: One-step protocol for nuclear intracellular proteins.
