Product Specific Information

Store vial at 4°C prior to restoration. For extended storage, aliquot contents and freeze at -20°C or below. Avoid freeze/thaw cycles. Centrifuge product if not completely clear after standing at room temperature. This product is stable for several weeks at 4°C as an undiluted liquid. Dilute only prior to immediate use.

This antibody is directed against the 6X His motif and is useful in determining its presence in various assays. It detects overexpressed proteins containing the 6X His epitope tag and has reacted with all His-tagged proteins tested. In western blotting of bacterial extracts, it does not cross-react with endogenous proteins. The antibody recognizes the His-tag (His-His-His-His-His-His) fused to either the amino- or carboxy-termini of targeted proteins in transfected or transformed cells.

Target Information

The 6X His tag is a synthetic oligopeptide consisting of six consecutive histidine residues (HHHHHH). It is commonly expressed at either the N- or C-terminal regions of recombinant proteins to facilitate isolation or purification via immobilized metal affinity chromatography. Epitope tagging allows detection of recombinant proteins even when no specific antibody exists. His-tagged proteins bind strongly to Ni²⁺ ions, enabling purification to homogeneity using Ni²⁺-NTA affinity chromatography. His tag-specific antibodies, such as this one, are valuable for detection or co-immunoprecipitation of tagged proteins.
