The MA1068 anti-IL17A antibody (clone 3-36.2.7.3) has successfully been paired as the coating antibody in a sandwich ELISA with detection antibody MA1069B (biotinylated conjugate of clone 3-41.2.6.1).
Typical dilutions for sandwich ELISA include 1 µg/mL for coating and 0.125–0.25 µg/mL for detection.
Target Information
Interleukin-17A (IL-17A, CTLA-8) is a CD4+ T cell-derived cytokine that promotes inflammatory responses in cell lines and is elevated in rheumatoid arthritis, asthma, multiple sclerosis, psoriasis, and transplant rejection.
IL-17A is a 32 kDa, disulfide-linked homodimer consisting of 136 amino acids and belongs to a six-member family (IL-17A–17F), signaling through the IL-17 receptor (IL-17R/CDw217).
High levels of IL-17A are produced by activated peripheral blood CD4+ T-cells. It enhances ICAM-1 expression in human fibroblasts and stimulates epithelial, endothelial, or fibroblastic cells to secrete IL-6, IL-8, G-CSF, and PGE2.
In the presence of IL-17A, fibroblasts support CD34+ hematopoietic progenitor proliferation and neutrophil maturation. Mouse, rat, and human IL-17A can induce IL-6 secretion in mouse stromal cells, indicating cross-species receptor recognition.
IL-17A regulates NF-kappa B and MAP kinase activities, stimulates IL-6 and COX-2 expression, and enhances nitric oxide (NO) production.
