Product Specific Information

• Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.

Target Information

Cytokeratins (CK) are intermediate filaments of epithelial cells, present in both keratinizing tissues (e.g., skin) and non-keratinizing cells (e.g., mesothelial cells). Although not traditional endothelial markers, cytokeratins are also found in some microvascular endothelial cells.
At least 20 different cytokeratins (CK), with molecular weights of 40–70 kDa and isoelectric points of 5–8.5, can be identified by two-dimensional gel electrophoresis.
They are classified into two types:

• Type I: Acidic polypeptides
• Type II: Basic polypeptides

Each epithelial cell expresses at least one member from each class. Monoclonal antibodies against cytokeratin proteins serve as useful markers for tumor identification and classification.
