• Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature (RT).
• Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95°C, followed by cooling at RT for 20 min.

Target Information

Cytokeratins (CK) are intermediate filaments of epithelial cells, found in both keratinizing (e.g., skin) and non-keratinizing tissues (e.g., mesothelial cells). Although not typical markers for endothelial cells, some microvascular endothelial cells also express cytokeratins.
At least 20 different cytokeratins (CKs) exist, ranging from 40–70 kDa with isoelectric points of 5–8.5. These can be identified via two-dimensional gel electrophoresis.
Cytokeratins are classified into two main types:

• Type I: Acidic polypeptides
• Type II: Basic polypeptides
  Each epithelial cell expresses at least one member from both families. Monoclonal antibodies to cytokeratin proteins are useful for tumor identification and classification.
