Target Information

MAP4K3 (also known as GLK) is a member of the STE20 family of serine/threonine protein kinases with an N-terminal catalytic domain and a C-terminal regulatory domain. It acts upstream of MEKK1 and is stimulated by UV radiation and TNF-alpha, leading to activation of the JNK signaling pathway.
MAP4K3 can directly activate PKC-θ during TCR signaling, and elevated MAP4K3 expression has been observed in T cells from patients with systemic lupus erythematosus. It functions as a pro-apoptotic kinase by activating BAX through modulation of PUMA, BAD, and BIM. The kinase activity is stimulated by UV radiation and TNF-alpha and specifically activates the c-Jun N-terminal kinase (JNK) pathway.
Previously known as RAB8-interacting protein-like 1 (RAB8IPL1), MAP4K3 has been renamed mitogen-activated protein kinase kinase kinase 3 (MAP4K3).
