Target Information
TBK1 (TANK-binding kinase 1) is a serine/threonine kinase belonging to the IKK family, involved in NF-κB activation and IRF3 signaling.
It acts as an essential mediator of gene expression during immune and inflammatory responses. NF-κB regulates numerous genes in response to extracellular stimuli and is inhibited in the cytoplasm by IKB proteins. Phosphorylation of IKB by the IKK complex leads to its degradation and subsequent NF-κB activation.
The IKK complex includes IKKa, IKKβ, and IKKK. TBK1 (also known as NAK or T2K) directly phosphorylates and activates IKKβ, mediating NF-κB activation in response to growth factors and PMA. TBK1 functions upstream of NIK and the IKK complex and plays a critical role in protecting embryonic liver from apoptosis.
