Antibody detects endogenous levels of TAK1 only when phosphorylated at Threonine 187.
Target Information
TAK1 (also known as MAP3K7) is a serine/threonine kinase involved in multiple signaling pathways. It was initially identified as a TGF-beta-activated kinase that interacts with TGF-beta receptor and TRAF6 to modulate activation of JNK and p38. TAK1 also participates in signaling through Wnt, BMP, activin, thyroid hormone, osmotic stress, endothelin, and ephrine pathways.
It regulates wound healing by promoting von Hippel-Lindau tumor suppressor expression and repressing PDGF-B and integrins β1/β5. TAK1 is essential for IKK activation in IL-1, IL-6, IL-18, TNF, CD40, TLR, and RIG-I signaling. Activation involves TAB2/3 binding and ubiquitination, followed by TAK1 autophosphorylation at Thr178, Thr184, Thr187, and Ser192, leading to IKK-β phosphorylation. TAK1 also represses human telomerase reverse transcriptase, indicating a role in cell lifespan regulation.
