Target Information
Anaplastic Lymphoma Receptor Tyrosine Kinase (ALK) belongs to the insulin receptor superfamily and is essential for brain development. Mutations, rearrangements, and amplifications in the ALK gene are implicated in various tumors, including anaplastic large cell lymphoma, neuroblastoma, and non-small cell lung cancer.
Chromosomal rearrangement typically produces a fusion gene combining ALK (chromosome 2) and NPM (chromosome 5), resulting in the oncogenic NPM-ALK fusion protein. ALK encodes a receptor tyrosine kinase with transmembrane and extracellular domains, which are absent in the NPM-ALK product. ALK shows high sequence similarity to LTK and plays a critical role in neuronal development and signaling.
