Target Information

Adenosine receptors (ARs) belong to the 7-transmembrane domain G-protein-coupled receptor superfamily. Structural, biochemical, and pharmacological analyses have identified four distinct AR subtypes (A1, A2a, A2b, A3). Activation of ARs mediates receptor subtype-specific physiological processes, including cardiac rate, smooth muscle tone, platelet aggregation, inflammation, cell growth and death, and neurotransmission.

The A1AR subtype is a glycoprotein that activates Gi and Go proteins in vitro. In intact cells, agonist binding to A1AR causes pertussis toxin-sensitive inhibition of adenylyl cyclase activity and may stimulate phospholipase C, leading to intracellular calcium mobilization. Activation of potassium channels by A1AR significantly affects cardiovascular function.

A1AR is highly expressed in the brain (notably cerebellum, hippocampus, thalamus, and cortex) and spinal cord, where it modulates neurotransmitter release. In white adipocytes, A1AR inhibits lipolysis and stimulates glucose uptake. It is also expressed in kidney and testis.
