Product Specific Information

The B-Ly7 monoclonal antibody reacts with human CD103, the alpha E integrin. CD103 non-covalently associates with integrin beta 7 and is expressed mainly on intraepithelial lymphocytes and a small subset of peripheral lymphocytes. It is also expressed in hairy cell leukemia (HCL) and some chronic B-cell lymphocytic leukemias.
In vitro stimulation of human T cells with mitogens induces upregulation of CD103. E-cadherin binds to CD103 and mediates lymphocyte homing to intestinal epithelium.

Each product contains:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and tested on PHA-activated human peripheral blood cells (5 µL per test, 100 µL final volume, 10⁵–10⁸ cells/test).

Important Notes:

• NovaFluor dyes are not compatible with DNA intercalating viability dyes (e.g., Propidium Iodide, 7-AAD, DAPI).
• Use Invitrogen LIVE/DEAD Fixable Dead Cell stains for viability assessment.
• CellBlox Plus Blocking Buffer (Cat. No. C001T06F01) is required for optimal staining and should be added to antibody cocktails prior to cell incubation (5 µL per sample).
• For single-color controls, use 5 µL of CellBlox Plus Blocking Buffer per 100 µL cell sample.

Target Information

CD103 (ITGAE, Integrin alpha E) is expressed on mucosa-associated T lymphocytes and activated TGF-β1 cells. It consists of a 150 kDa alpha chain and a 120 kDa beta chain. Expressed by 0.5–2% of resting lymphocytes in peripheral blood and lymphoid organs. It binds E-cadherin via integrin β7 forming human mucosal lymphocyte-1 antigen.
CD103 plays a role in adhesion and activation of intestinal intraepithelial lymphocytes (IEL).
Associated diseases include hairy cell leukemia and splenic marginal zone lymphoma.
