Product Specific Information
The 53-6.7 monoclonal antibody reacts with the mouse CD8a molecule (approx. 32–34 kDa). CD8a is expressed as a heterodimer with CD8β (CD8αβ) or as a homodimer (CD8αα). Thymocytes and αβ TCR T cells express CD8αβ, while γδ TCR T cells, intestinal intraepithelial lymphocytes, and dendritic cells express CD8αα. CD8 binds MHC class I and associates with p56lck, playing a role in T cell development and activation.
Applications Reported: Flow cytometric analysis.
Applications Tested: Tested by flow cytometric analysis of mouse thymocyte or splenocyte suspensions. Recommended ≤0.5 µg/test (100 µL final volume). Cell number: 10⁵–10⁸ cells/test. Titrate for optimal performance.

Target Information
Cluster of differentiation 8 (CD8) is a type I transmembrane glycoprotein of the immunoglobulin family. It plays a key role in signal transduction, T cell differentiation, and activation. CD8 is expressed on T cells as a CD8αβ heterodimer (co-receptor for MHC-I) and as CD8αα homodimers on subsets of T cells, NK cells, monocytes, and dendritic cells. Engagement of MHC-I/peptide complexes triggers Lck recruitment, leading to cytokine production and cytotoxic T lymphocyte (CTL) activation. CTLs are essential for pathogen and tumor cell clearance. Differentiation of naive CD8+ T cells into CTLs is enhanced by IL-2, IL-12, and TGF-β1.
