Product Specific Information
This MFRDTRK monoclonal antibody recognizes human and mouse IkB alpha, a member of the inhibitor family of NF-kappaB signaling. IkB alpha prevents NF-kappaB nuclear translocation by masking its nuclear localization signals, keeping it inactive in the cytoplasm. Upon stimulation (e.g., TNF alpha, IL-1, ROS), IkB kinase (IKK) phosphorylates IkB alpha, leading to ubiquitination and degradation, allowing NF-kappaB activation.
Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated for intracellular staining of normal human peripheral blood cells at 5 µL (0.125 µg) per test.
Recommended Cell Number: 10⁵–10⁸ cells/test.

Target Information
IkB-alpha (≈40 kDa) inhibits NF-kappaB activity by binding NF-kappaB dimers in the cytosol and masking their nuclear localization sequence. Degradation of IkB-alpha, triggered by stimuli such as TNF-alpha, IL-1 beta, LPS, UV, and viral infection, releases NF-kappaB for nuclear translocation and gene activation.
The human IkB-alpha gene is located on chromosome 14q. Phosphorylation on serine residues 32 and 36 by IKK alpha (CHUK) induces degradation, enabling NF-kappaB activation. The IKK complex (IKK alpha, IKK beta, IKK gamma) is essential for this process.
