Target Information

COX2 converts arachidonate to prostaglandin H2 (PGH2), a key step in prostanoid synthesis including inflammatory prostaglandins. This reaction involves:

1. Cyclooxygenase (COX) reaction: Conversion of arachidonate to prostaglandin G2 (PGG2)
2. Peroxidase reaction: Reduction of PGG2 to PGH2

COX2 is constitutively expressed in tissues such as endothelium, kidney, and brain, and is up-regulated in pathological conditions like cancer and inflammation.
Up-regulation is linked to increased cell adhesion, phenotypic changes, resistance to apoptosis, and tumor angiogenesis.
In cancer cells, COX2 promotes production of prostaglandin E2 (PGE2), influencing motility, proliferation, and apoptosis resistance.
COX2 activity is naturally inhibited by calcitriol (active Vitamin D) and suppressed by glucocorticoids, which interfere with PTGS2 gene transcription.
It is also a target of NSAIDs such as aspirin, which reduce inflammation-related pain and swelling.
