Product Specific Information
For reconstitution, add 100 µL of distilled water to obtain a final antibody concentration of approximately 1 mg/mL.
For conjugation experiments, perform an additional desalting step (e.g., Zeba Spin Desalting Columns, 7K MWCO, 0.5 mL, Product #89882) to ensure carrier-free conditions.
Target Information
In the intact cell, DNA closely associates with histones and other nuclear proteins to form chromatin. Chromatin remodeling is essential for transcriptional regulation, with histone acetylation being a major contributor. Acetylation of lysine residues in histone tails induces conformational changes, enhancing DNA accessibility to transcription factors.
Several mammalian proteins act as nuclear histone acetylases, including GCN5, PCAF, p300/CBP, HAT1, and TAF II p250. Conversely, histone deacetylation is linked to transcriptional silencing. The histone deacetylase (HDAC) family includes HDAC1–9.
HDAC9 and HDAC9a are alternatively spliced isoforms; HDAC9a is 132 amino acids shorter but retains the catalytic domain and deacetylase activity. Both isoforms repress myocyte enhancer-binding factor 2-mediated transcription and are expressed in brain, skeletal muscle, kidney, placenta, and pancreas.
