DNA topoisomerases are nuclear enzymes that regulate the topological structure of DNA in eukaryotic cells by transiently breaking and rejoining DNA strands. They play essential roles in DNA replication, recombination, and transcription, and are targets of many anticancer drugs. Mitochondrial Topo I (DNA topoisomerase I, mitochondrial) is a 601 amino acid protein that relieves DNA strain during mitochondrial DNA replication. As a type IB topoisomerase, it requires divalent metals (Ca²⁺ or Mg²⁺) and an alkaline pH for optimal activity.
Target Information
This gene encodes a mitochondrial DNA topoisomerase involved in modifying DNA topology. The encoded protein, a type IB topoisomerase, catalyzes transient DNA strand breaks and rejoining to relieve supercoiling during mitochondrial DNA replication and transcription. Multiple alternatively spliced transcript variants encoding different isoforms have been identified.
