Product Specific Information

The M1/69 monoclonal antibody reacts with the mouse CD24 molecule, also known as Heat Stable Antigen (HSA). This 35–50 kDa molecule is anchored in the plasma membrane via phosphatidylinositol and is expressed by erythrocytes, thymocytes, peripheral lymphocytes, and myeloid lineage cells. CD24 is variably glycosylated, leading to heterogeneity in molecular mass across cell types. Antibodies to CD24 show subtle differences in staining levels among lymphocyte populations. CD24 expression has been used to identify stages of B lymphopoiesis in mouse bone marrow. It has been reported that P-selectin (CD62P) binds to CD24.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes; ≤1.0 µg/test recommended.
A test is defined as the amount of antibody required to stain a 100 µL cell sample. Cell number can vary (10⁵–10⁸ cells/test). Titrate for optimal results.

Fluorochrome Information:
Brilliant Ultra Violet™ 496 (BUV496) emits at 496 nm and is intended for use on cytometers with an ultraviolet (355 nm) laser. Ensure the instrument supports this fluorochrome.

Staining Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies (e.g., Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (#SB-4401-42) or Brilliant Stain Buffer™ (#00-4409-75) to minimize nonspecific interactions.

Light Sensitivity:
Protect from light; BUV496 is sensitive to photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (#00-8222-49) or 1-step Fix/Lyse Solution (#00-5333-54) for up to 3 days at 4°C in the dark with minimal brightness loss. Clone-specific performance may vary.

Target Information

CD24 is a GPI-anchored glycoprotein expressed at multiple stages of B-cell development, from pro-B to mature B-cells, but low or absent in plasma cells. It is also expressed in most B-lineage acute lymphoblastic leukemias, B-cell CCLs, and B-cell non-Hodgkin’s lymphomas. CD24 may regulate B-cell proliferation, maturation, and autoimmunity control.
