Product Specific Information

Description:
The WM-53 monoclonal antibody reacts with human CD33, also known as GP67 and P67, a 67 kDa type I transmembrane glycoprotein belonging to the Siglec (sialic acid-binding Ig superfamily lectin) family. It is highly specific to the hematopoietic compartment and is expressed on monocytes, activated T cells, granulocytes, myeloid progenitors, and mast cells.

Applications Reported:
This WM-53 (WM53) antibody is reported for use in flow cytometric analysis.

Applications Tested:
Pre-diluted and tested by flow cytometric analysis of normal human peripheral blood cells. Recommended usage is 5 µL (0.5 µg) per test (final volume 100 µL). Cell number should be determined empirically (10⁵–10⁸ cells/test).

Fluorochrome Information:
Brilliant Ultra Violet™ 496 (BUV496) emits at 496 nm and is intended for use on cytometers equipped with an ultraviolet (355 nm) laser. Ensure instrument compatibility before use.

Staining Buffer Recommendation:
When using multiple polymer dye-conjugated antibodies, use:

• Super Bright Complete Staining Buffer (Product # SB-4401-42) or
• Brilliant Stain Buffer™ (Product # 00-4409-75)
  to minimize non-specific polymer interactions.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect vial and stained samples from light.

Fixation:
Samples can be stored in:

• IC Fixation Buffer (Product # 00-8222-49)
• 1-step Fix/Lyse Solution (Product # 00-5333-54)
  for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

CD33 is a transmembrane protein of the Siglec (sialic acid-binding immunoglobulin-like lectin) family. It contains immunoreceptor tyrosine-based inhibitory motifs (ITIMs) that recruit SHP-1 and SHP-2 phosphatases for signaling and receptor regulation. CD33 is expressed on myelomonocytic lineage cells, binds sialic acid residues on cell surfaces, and serves as a therapeutic target for acute myeloid leukemia. It is found on granulocyte and macrophage precursors in bone marrow but not on pluripotent stem cells, and is a useful marker for distinguishing myelogenous leukemia from lymphoid or erythroid types. Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
