Product Specific Information

Description:
The eBio1D3 monoclonal antibody reacts with mouse CD19, a 95 kDa transmembrane glycoprotein expressed by B cells during all stages of development except terminally differentiated plasma cells. Follicular dendritic cells also express CD19. CD19 forms a multimolecular complex with CD21, CD81, and MHC class II that associates with the BCR, inducing tyrosine phosphorylation, calcium flux, and B cell proliferation.

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Tested on mouse splenocytes by flow cytometry at ≤0.5 µg/test. One test is defined as the amount of antibody required to stain a 100 µL cell sample (10⁵–10⁸ cells/test). Optimal dilution should be determined empirically.

Fluorochrome Information:
Brilliant Ultra Violet™ 496 (BUV496) emits at 496 nm and is intended for use on cytometers equipped with a 355 nm ultraviolet laser. Ensure instrument compatibility before use.

Staining Recommendations:
When using multiple polymer dye-conjugated antibodies (Super Bright™, Brilliant Violet™, Brilliant Ultra Violet™), use Super Bright Complete Staining Buffer (Product # SB-4401-42) or Brilliant Stain Buffer™ (Product # 00-4409-75) to minimize non-specific interactions.

Light Sensitivity:
This dye is sensitive to photo-induced oxidation. Protect vials and stained samples from light.

Fixation:
Samples may be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-54) for up to 3 days at 4°C in the dark with minimal loss of signal.

Target Information

CD19 is a member of the immunoglobulin superfamily with two Ig-like domains, expressed on all peripheral B cells. It is also found on certain myeloid leukemia cells. CD19 functions as a signal-amplifying co-receptor for the B cell receptor (BCR) and can signal independently, playing a central role in B cell activation and regulation. Mutations in CD19 can result in hypogammaglobulinemia, while overexpression leads to B cell hyperactivity.
