Product Specific Information

The C17.8 antibody reacts with the p40 subunit of IL-12 and IL-23. It neutralizes IL-12 and IL-23 bioactivity.
For specific neutralization of IL-23 only, see clone G23-8 (anti-mouse IL-23 p19).

IL-12 is a 70 kDa heterodimeric cytokine composed of p40 and p35 subunits, secreted by activated monocytes, macrophages, and dendritic cells.
IL-23 is a heterodimer composed of p19 and p40 proteins, expressed by activated dendritic cells.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Intracellular staining and flow cytometric analysis of stimulated mouse thioglycolate-elicited peritoneal exudate cells.
Recommended usage ≤ 0.25 µg/test (100 µL final volume). Optimal titration required.

eFluor™ 450 is an alternative to Pacific Blue™, excited at 405 nm and emits at 445 nm.
Ensure instrument compatibility with violet laser detection.

Filtration: 0.2 µm post-manufacturing filtered.

Target Information

IL-12 is produced by dendritic cells, neutrophils, macrophages, and lymphoblastoid cells in response to antigenic stimulation.
It is a heterodimer encoded by IL-12A (p35) and IL-12B (p40).
IL-23 shares the IL-12B subunit and includes IL-23A (p19).

Functional Roles:

• IL-12 increases endothelial adhesion molecule expression and promotes leukocyte migration toward tumors.
• IL-23 stimulates memory T-cells and supports Th17 cell functions, enhancing neutrophil chemokine and pro-inflammatory cytokine production.
