Product Specific Information

The C8.6 antibody reacts with the p40 subunit of human IL-12 p70 and IL-23.
IL-12 is a 70 kDa heterodimeric cytokine composed of p40 and p35 chains, secreted primarily by activated monocytes, macrophages, and dendritic cells.
IL-23 is a related heterodimeric cytokine composed of disulfide-linked p40 and p19 subunits.

Applications Reported:

• Intracellular staining followed by flow cytometric analysis.

Applications Tested:

• Pre-titrated and tested for intracellular staining and flow cytometry of stimulated human peripheral blood cells.
• Recommended usage: 5 µL (0.25 µg) per test (100 µL final volume, 10⁵–10⁸ cells/test).

Fluorochrome Information:

• eFluor® 450 is an alternative to Pacific Blue®.
• Excitation: 405 nm; Emission: 445 nm; Laser: Violet.

Target Information

IL-12:
Produced by dendritic cells, neutrophils, macrophages, and lymphoblastoid cells upon antigenic stimulation.
Encoded by IL-12A (p35) and IL-12B (p40) genes.
Enhances vascular endothelial adhesion molecule-1 expression, directs leukocytes toward tumors, and facilitates tumor necrosis.

IL-23:
Shares IL-12B (p40) with IL-12 and includes IL-23A (p19) subunit.
Stimulates memory T-cells and Th17 cell functions, promoting neutrophil chemokine and pro-inflammatory cytokine expression.
