The monoclonal antibody 2F12 recognizes human, mouse, and rat ITK (Interleukin-2 inducible T-cell kinase). ITK is a member of the Tec family of kinases (including Tec, Btk, Bmx, and Txk) that play crucial roles in activation of phospholipase C and calcium-dependent signaling following antigen receptor stimulation.

ITK is highly expressed in T cells and is required for optimal TCR-induced PLCγ1 activation. Mice lacking ITK show significant defects in T cell development and signaling. Mutations in ITK are associated with lymphoproliferative syndrome EBV-associated autosomal type 1 (LPSA1). ITK also contributes to T cell-mediated inflammatory disorders such as atopic dermatitis, psoriasis, and allergic asthma.

Applications Reported:
This 2F12 antibody has been reported for use in western blotting, immunohistochemical staining of formalin-fixed paraffin-embedded tissue sections, and microscopy.

Applications Tested:
Tested by immunohistochemistry of formalin-fixed paraffin-embedded human tissue using high pH antigen retrieval. Recommended concentration ≤ 1 µg/mL. Optimal dilution should be determined experimentally.

Purity: Greater than 90%, as determined by SDS-PAGE.
Aggregation: Less than 10%, as determined by HPLC.
Filtration: 0.2 µm post-manufacturing filtered.

Target Information

ITK is a tyrosine kinase expressed in T cells containing both SH2 and SH3 domains. It is involved in T-cell proliferation and differentiation. The Tec family of non-receptor tyrosine kinases includes Tec, Itk (Emt/Tsk), Btk, Bmx, Txk (Rlk), and Dsrc28C. All members have SH3 and SH2 domains; most also contain PH and TH domains at their amino termini. ITK associates with CD28 and becomes activated upon CD28 ligation.
