The eBio5E8-C7-F10 monoclonal antibody reacts with human CD182 (CXCR2, IL-8Rβ). CD182 is a 67–70 kDa member of the seven-transmembrane G-protein coupled receptor (GPCR) family. It is expressed as a homodimer or heterodimer with CD181 (CXCR1, IL-8Rα) on granulocytes, NK cells, subsets of T cells, mast cells, monocytes, endothelial cells, megakaryocytes, and oligodendrocytes.

Binding of its ligands — including IL-8, NAP-2, GCP-2, and GRO-α/β/γ — induces biological responses such as cell activation, chemotaxis, proliferation, and angiogenesis. While both CD181 and CD182 mediate chemotaxis and calcium signaling, only CD181 triggers phospholipase D activation and respiratory burst. IL-8 predominantly affects neutrophil function via CD181.

This antibody is reported for use in flow cytometric analysis.

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood. Recommended ≤0.5 µg per test (defined as antibody amount in 100 µL final volume). Cell number should be empirically determined (10⁵–10⁸ cells/test). Titrate for optimal performance.

Target Information

CXCR2 is a receptor for interleukin-8 (IL-8), a potent neutrophil chemotactic factor. It belongs to the GPCR family. IL-8 binding activates neutrophils via a G-protein-mediated phosphatidylinositol-calcium second messenger system. CXCR2 binds IL-8, GRO/MGSA, and NAP-2 with high affinity and is expressed in various tissues, including placenta and thymus (based on EST data).
