Description:
The MHR73-11 monoclonal antibody reacts with human CD180 (RP105), a 105 kDa type I transmembrane molecule and member of the TLR family. It associates with MD-1 and is expressed on B cells, monocytes/macrophages, and dendritic cells. RP105/MD-1 complex mediates B cell recognition and signaling of LPS, leading to B cell activation, increased CD80 expression, and DNA synthesis. RP105 ligation protects B cells from apoptosis induced by irradiation or dexamethasone. Loss of RP105 expression is associated with B cell activation and increased disease activity in SLE patients.

Applications Reported:
MHR73-11 is reported for use in flow cytometry, immunoprecipitation, immunohistochemistry, and in vitro functional assays. For functional assays, use the Functional Grade purified MHR73-11 (Product #16-1809-82).

Applications Tested:
Tested by flow cytometric analysis of normal human peripheral blood cells. Recommended ≤1 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize dilution for each assay.

Target Information

CD180 (RP105, Bgp95, LY64) is a type I membrane glycoprotein of the Toll-like receptor (TLR) family. It has a short cytoplasmic tail lacking the TIR domain and requires coexpression with MD-1. CD180 regulates LPS recognition and B cell signaling through interaction with the TLR4 complex. Ligation by monoclonal antibodies induces B cell activation and upregulation of CD80/CD86. CD180 contains extracellular leucine-rich repeats (LRR) and a short cytoplasmic tail. It is associated with diseases such as Kawasaki Disease and rheumatic disorders.
