7D4 recognizes mouse CD25 at an epitope comprising amino acids 184–194 (REHHRFLASEE).
This epitope is distinct from the IL-2 binding site and does not block IL-2 binding to CD25.
Murine CD25 is a 55 kDa interleukin-2 receptor alpha chain (IL-2Rα), expressed by early progenitors of T and B lineage as well as activated mature T and B lymphocytes.
CD25 alone binds IL-2 with low affinity, but associates with CD122 (IL-2Rβ) and CD132 (common γ chain) to form the high-affinity IL-2 receptor.

Target Information

CD25 (IL2 receptor alpha chain / IL2RA) is a cytokine receptor involved in T and B lymphocyte proliferation.
The IL2 receptor complex is heterotrimeric, composed of IL2RA, IL2RB, and IL2RG.

• Homodimeric IL2RA → low-affinity receptor
• Homodimeric IL2RB → medium-affinity receptor

IL2 expression in mature thymocytes is monoallelic, representing a unique regulatory mechanism.
IL2 is mainly produced by mature T cells and acts as a growth, differentiation, and apoptosis regulator.
It stimulates B cell proliferation, enhances NK cell activity, and inhibits granulocyte macrophage colony formation.
Disruption of the IL2RA gene in mice leads to ulcerative colitis-like disease, indicating its role in immune response regulation.
Mutations in IL2RA are associated with interleukin 2 receptor alpha deficiency.
