Target Information

Nerve growth factor (NGF) influences the survival and differentiation of various neuronal and non-neuronal cells. NGF binds to specific cell surface receptors with high and low affinity, with only the high-affinity receptor mediating signal transduction. Cross-linking studies with ¹²⁵I-NGF have shown that the high-affinity receptor in rat PC12 cells forms an Mr 158,000 complex with NGF, while the low-affinity receptor forms an Mr 100,000 complex.
Full-length cDNA clones for both human and rat NGF receptor genes show greater than 90% homology. NGF regulates growth and differentiation of sympathetic and sensory neurons, and mutations in the NGF gene are associated with hereditary sensory and autonomic neuropathy type 5 (HSAN5). Dysregulation of NGF expression is linked to allergic rhinitis.
