Product Specific Information

The UCHT1 monoclonal antibody reacts with human CD3e, a 20 kDa subunit of the TCR complex. CD3e, along with CD3γ and CD3δ, is essential for proper TCR assembly and surface expression. CD3 is expressed on thymocytes and all mature T cells. Crosslinking of TCR via immobilized UCHT1 triggers intracellular signaling leading to activation and proliferation.

Each product includes:

• 1 vial of NovaFluor conjugate
• 1 vial of CellBlox Plus Blocking Buffer

Applications Reported: Flow cytometric analysis
Applications Tested: Pre-titrated and validated for flow cytometry using normal human peripheral blood cells. Recommended use: 5 µL (0.4 µg)/test in 100 µL total volume. Cell number: 10⁵–10⁸ cells/test (empirically determined).

Note:
NovaFluor dyes are not compatible with DNA intercalating viability dyes such as propidium iodide, 7-AAD, or DAPI. Use Invitrogen LIVE/DEAD Fixable Dead Cell stains instead.

This product ships with CellBlox Plus Blocking Buffer (Cat. No. C001T06F01), which provides improved formulation. Use 5 µL per sample for optimal staining. Add CellBlox Plus Blocking Buffer to antibody cocktails before combining with cells.

NovaFluor conjugates utilize Phiton technology, based on nucleic acid dye structures engineered for optimized fluorescence with minimal spillover and spread.

Target Information

The CD3 complex is essential for antigen-recognition signal transduction and regulation of TCR surface expression. It includes CD3γ, CD3δ, CD3ε, and CD3ζ subunits, encoded on chromosome 11. These subunits interact with intracellular signaling molecules via tyrosine-based motifs. CD3 is expressed on 68–82% of peripheral T lymphocytes, 65–85% of thymocytes, and Purkinje cells, but absent on B and NK cells. CD3 gene defects are associated with immunodeficiency.
