Product Specific Information

This NLA14 monoclonal antibody recognizes mouse VSIG4 (V-Set and Immunoglobulin domain containing 4), also known as CRIg or Z39Ig.
VSIG4 is a type I transmembrane glycoprotein related to the B7 family and expressed exclusively on tissue-resident and tumor-infiltrating macrophages. It binds complement components C3b and iC3b, inhibiting the alternative complement pathway and enhancing phagocytosis of complement-opsonized pathogens. VSIG4 also suppresses T cell activation, proliferation, and IL-2 production, contributing to immune tolerance and inflammation suppression.

Expression of VSIG4 on tumor-infiltrating macrophages suggests its role in immune evasion. Pro-inflammatory stimuli such as TNF and LPS down-regulate VSIG4 expression.
Peritoneal macrophages in Balb/c mice express higher levels of VSIG4 than in C57Bl/6 or Swiss Webster mice.

Applications Tested: Tested on resident peritoneal macrophages; recommended ≤0.5 µg/test in 100 µL final volume (10⁵–10⁸ cells/test). Optimize titration as needed.

Fluorochrome Information:
PE-eFluor 610 can be excited at 488–561 nm and emits at 607 nm. Recommended filter: 610/20 band pass (PE-Texas Red equivalent).
Ensure instrument compatibility.

Light Sensitivity: Protect from light due to photo-induced oxidation.
Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-57) with minimal impact on brightness and FRET efficiency.
