Product Specific Information

The monoclonal antibody 20Raj1 recognizes the human Ki-67 protein, which exists in two isoforms (345 and 395 kDa) expressed in dividing cells. Ki-67 is detected during active phases of the cell cycle (G1, S, G2, M) but absent in resting cells (G0). During interphase, expression is nuclear; during mitosis, it redistributes to chromosomes and associates with heterochromatin-bound proteins such as CBX3.

This antibody also recognizes canine Ki-67.

Applications Reported: Intracellular staining followed by flow cytometric analysis.
Applications Tested: Pre-titrated and tested by intracellular staining of normal human peripheral blood cells. Recommended use: 5 µL (0.25 µg) per test (final volume 100 µL; 10⁵–10⁸ cells/test).

Fluorophore Information:
PE-eFluor® 610 can be excited with 488–561 nm lasers and emits at 607 nm. Recommended filter: 610/20 band pass (PE-Texas Red® equivalent). Protect from light—tandem dye is sensitive to photo-induced oxidation.

Fixation:
Samples can be stored in IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency.

Target Information

Ki-67 is a nuclear protein expressed during late G1, S, G2, and M phases. It associates with euchromatin and heterochromatin via its FHA domain, showing cell cycle–dependent localization. Ki-67 is widely used as a proliferation marker, absent in G0 cells but increasing through mitosis. It is valuable for assessing tumor cell proliferation and correlates strongly with histological grade. Also used as a neuronal marker for cell cycling and proliferation.
