Product Specific Information

The A20 monoclonal antibody reacts with the mouse CD45 molecule (leukocyte common antigen, LCA) in CD45.1-expressing mouse strains such as SJL/J, DA, STS/A, and RIII. CD45.1 is expressed by all leukocytes in these strains.

• Applications Reported: Flow cytometric analysis
• Applications Tested: Mouse splenocytes, ≤1 µg/test (100 µL final volume, 10⁵–10⁸ cells/test). Optimize titration for best results.

Super Bright 645 is a tandem dye excited by the violet laser (405 nm) and emits at 645 nm. Recommended filter: 660/20 bandpass. Ensure instrument compatibility.

When using multiple Super Bright dyes: Use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize polymer interactions. Protect from light due to photo-induced oxidation sensitivity.

Fixation: Samples can be stored up to 3 days at 4°C in the dark using IC Fixation Buffer (Product # 00-8222) or 1-step Fix/Lyse Solution (Product # 00-5333) with minimal impact on brightness and FRET efficiency. Performance may vary by clone.

Target Information

CD45 (LCA, leukocyte common antigen) is a receptor-type protein tyrosine phosphatase expressed on all nucleated hematopoietic cells, constituting ~10% of lymphocyte surface proteins. It plays key roles in lymphocyte development and antigen signaling, regulating Src-family kinases.
CD45 exists as multiple isoforms due to alternative splicing, differing in extracellular domains but sharing transmembrane and cytoplasmic regions. Isoform expression varies by cell type and physiological state.
Beyond immunoreceptor signaling, CD45 supports cell survival via integrin-mediated pathways and participates in DNA fragmentation during apoptosis.
CD45RA is one isoform with restricted expression among lymphoid subtypes.
