Product Specific Information

  • Immunohistochemistry (PFA fixed): incubate antibody for 30 min at room temperature (RT).
  • Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA, pH 9.0, for 45 min at 95 °C, followed by cooling at RT for 20 min.

Target Information

Ki-67 is a nuclear protein expressed during various stages of the cell cycle, particularly in late G1, S, G2, and M phases. It contains a forkhead-associated (FHA) domain that mediates binding to euchromatin at the perichromosomal layer, centromeric heterochromatin, and the nucleolus. Ki-67 shows cell cycle–dependent localization: perinucleolar expression at G1, nuclear matrix localization at G2, and chromosomal association during mitosis.
Because Ki-67 is absent in G0 cells but increases from G1 through mitosis, it serves as a reliable proliferation marker. Ki-67 antibodies are used to determine the growth fraction in neoplasms, with prognostic value comparable to the tritiated thymidine-labeling index. A low Ki-67 index correlates strongly with histologically low-grade tumors. It is also widely used as a neuronal marker for cell cycling and proliferation.
