  • Immunohistochemistry (PFA fixed): Incubate antibody for 30 min at room temperature.
  • For formalin-fixed tissues: Heat tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 min at 95°C, then cool at room temperature for 20 min.

Target Information

Ki-67 is a nuclear protein expressed during various stages of the cell cycle, particularly in late G1, S, G2, and M phases. It contains a forkhead-associated (FHA) domain that mediates association with euchromatin at the perichromosomal layer, centromeric heterochromatin, and nucleolus.
Expression patterns vary by cell cycle stage — perinucleolar in G1, nuclear matrix in G2, and chromosomal during M phase.
Ki-67 serves as a proliferation marker, absent in G0 cells but increasing from G1 through mitosis. It is widely used to assess the growth fraction in neoplasms, correlating with tumor grade and prognosis.
