Product Specific Information
Immunohistochemistry (PFA fixed): Incubate antibody for 30 minutes at room temperature.
Staining of formalin-fixed tissues requires heating tissue sections in 10 mM Tris with 1 mM EDTA (pH 9.0) for 45 minutes at 95°C, followed by cooling at room temperature for 20 minutes.
Target Information
CD21 (complement receptor 2, CR2, C3D receptor, EBV receptor) binds C3 complement fragments, particularly breakdown fragments covalently attached to complement-activating surfaces or antigens.
It plays a critical role in:
- Uptake and retention of immunocomplexes
- Survival of memory B cells
- Development and maintenance of humoral immune responses to T-dependent antigens
CD21 also serves as a receptor for Epstein-Barr virus (EBV) and mediates prion targeting to follicular dendritic cells, accelerating neuroinvasion after peripheral exposure.
A soluble form (sCD21) is shed from lymphocytes and retains ligand-binding ability.
CD21 interacts with CD23 and forms a signaling complex with CD19, CD81, and Leu13, contributing to B cell activation.
Genetic variations in CD21 are associated with susceptibility to systemic lupus erythematosus type 9 (SLEB9).
Alternative splicing generates multiple isoforms of CD21.
