Target Information

Isocitrate dehydrogenases catalyze the oxidative decarboxylation of isocitrate to 2-oxoglutarate. These enzymes belong to two distinct subclasses, one utilizing NAD(+) and the other NADP(+) as electron acceptors.
Five isocitrate dehydrogenases have been reported: three NAD(+)-dependent enzymes localized to the mitochondrial matrix and two NADP(+)-dependent enzymes (one mitochondrial, one cytosolic).
NAD(+)-dependent isocitrate dehydrogenases catalyze the rate-limiting step of the tricarboxylic acid cycle. Each isozyme consists of two alpha subunits, one beta subunit, and one gamma subunit.
The IDH3G gene encodes the gamma subunit of one NAD(+)-dependent isozyme and is a candidate gene for periventricular heterotopia. Several alternatively spliced transcript variants have been described.
