Target Information
GCN1L1 acts as a positive activator of EIF2AK4/GCN2 protein kinase activity in response to amino acid starvation. It forms a complex with EIF2AK4/GCN2 on translating ribosomes and facilitates the delivery of uncharged tRNAs to the EIF2AK4/GCN2 tRNA-binding domain, stimulating kinase activity. This process contributes to repression of global protein synthesis and activation of gene-specific mRNA translation, such as ATF4, through EIF2AK4/GCN2-mediated phosphorylation of eIF2α (EIF2S1) on Ser-52, enabling ATF4-mediated reprogramming of amino acid biosynthetic gene expression to alleviate nutrient depletion.
