Target Information
HEF-1 is a multifunctional protein involved in integrin-based signaling that affects cell motility, growth, apoptosis, and oncogenic transformation.
The Cas family of docking proteins are substrates of focal adhesion kinase (FAK) and the Src family of tyrosine kinases—two key targets for drug development.
HEF1 overproduction increases expression of genes linked to motility, transformation, and invasiveness, including metalloproteinases, MLCK, p160ROCK, and ErbB1.
It also mediates apoptosis in epithelial-derived cell lines such as MCF7 and HeLa.
Overexpression of BCAR1 (p130Cas), a related protein, has been associated with tamoxifen resistance, underscoring the importance of this family in cancer prognosis.
