Product Specific Information

Description:
The 90 monoclonal antibody reacts with the mouse CD38 molecule, an approximately 42 kDa type II transmembrane protein. CD38 expression increases on B cells during differentiation, then down-regulates on germinal center B cells and mature plasma cells. It is also expressed on subsets of thymocytes, mature T cells, and NK cells. Crosslinking of CD38 on resting B cells induces proliferation, enhanced by IL-4 and LPS. CD38, a counter-receptor for CD31, functions as an ectoenzyme with cyclase and hydrolase activity, playing a role in lymphocyte activation and differentiation.

Reported for use in flow cytometric analysis.

Applications Tested:
Tested on mouse splenocytes for flow cytometric analysis. Recommended ≤0.125 µg/test (in 100 µL final volume). Optimal titration is advised. Cell number per test: 10⁵–10⁸.

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact. Clone-specific performance should be verified empirically.

Target Information

CD38 (NAD⁺ glycohydrolase):
A type II transmembrane glycoprotein that induces activation, proliferation, and differentiation of mature lymphocytes, and mediates apoptosis of progenitor cells. Functions as a multi-catalytic ectoenzyme (ADP-ribosyl cyclase, cyclic ADP-ribose hydrolase, NAD⁺ glycohydrolase) and as a cell surface receptor.
Antibodies to CD38 are useful for:

• Subtyping lymphomas and leukemias
• Detecting plasma cells (e.g., myelomas)
• Identifying activated B and T cells

CD38 is highly expressed in pancreas, liver, kidney, malignant lymphoma, and neuroblastoma.
Associated diseases include chronic lymphocytic leukemia and Richter’s Syndrome.
