Product Specific Information
The 4H11 monoclonal antibody reacts with human CD34, also known as mucosialin. CD34 belongs to a protein family including endoglycan and podocalyxin, characterized by single-pass transmembrane structure and glycosylated extracellular domains. CD34 is a well-established marker for hematopoietic progenitors and is also expressed on certain mesenchymal stem cells, tumor cell lines, and vascular endothelia.
Epitopes of CD34 are categorized into three classes (I–III) based on enzymatic sensitivity; 4H11 belongs to class III, recognizing a protein epitope.
Applications Reported: Flow cytometric analysis.
Applications Tested: Pre-titrated for flow cytometry using normal human peripheral blood cells at 5 µL (0.25 µg)/test (in 100 µL final volume).
Light Sensitivity: Protect from light due to photo-induced oxidation.
Fixation: Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark.
Excitation: 488–561 nm
Emission: 775 nm
Compatible Lasers: Blue, Green, Yellow-Green

Target Information
CD34 is a highly glycosylated monomeric surface protein (111–115 kDa) expressed on various stem cell populations. It serves as a marker for hematopoietic stem cells and may function as a receptor involved in adhesion, differentiation, and proliferation. CD34 possibly mediates stem cell attachment to bone marrow extracellular matrix or stromal cells and may act as a scaffold for glycans binding to lectins. The intracellular domain is a phosphorylation site for protein kinase C, suggesting a role in signal transduction.
