Product Specific Information

Description:
The WM-53 monoclonal antibody reacts with human CD33 (GP67, P67), a 67 kDa type I transmembrane glycoprotein in the Siglec family. CD33 is expressed in hematopoietic cells such as monocytes, activated T cells, granulocytes, myeloid progenitors, and mast cells.

Applications Reported:
Suitable for flow cytometric analysis.

Applications Tested:
Pre-titrated and validated for flow cytometric analysis of normal human peripheral blood cells. Recommended usage: 5 µL (0.25 µg) per test in 100 µL final volume. Cell count: 10⁵–10⁸ cells/test (empirically determined).

Light Sensitivity:
This tandem dye is sensitive to photo-induced oxidation. Protect from light.

Fixation:
Samples can be stored in IC Fixation Buffer (Product #00-8222-49) or 1-step Fix/Lyse Solution (Product #00-5333-54) for up to 3 days at 4°C in the dark with minimal impact on brightness and FRET efficiency. Clone-specific performance should be verified empirically.

Target Information

CD33 is a transmembrane protein of the Siglec (sialic acid-binding immunoglobulin-like lectin) family containing ITIM motifs that recruit SHP-1 and SHP-2 phosphatases. It mediates inhibitory signaling and receptor endocytosis. CD33 is expressed on myelomonocytic lineage cells, granulocyte and macrophage precursors, and peripheral monocytes, but not on pluripotent stem cells. It serves as a marker to distinguish myelogenous leukemia cells and is a therapeutic target for acute myeloid leukemia. Diseases associated with CD33 dysfunction include gallbladder lymphoma and extracutaneous mastocytoma.
