Product Specific Information

This TNKUPJ monoclonal antibody recognizes mouse CD163, a 130 kDa surface receptor expressed by specific macrophage subsets such as splenic red pulp macrophages, Kupffer cells, intestinal lamina propria macrophages, and peritoneal macrophages. Mouse monocytes do not express CD163, and it is not a reliable M2 macrophage marker in mice.

This antibody detects CD163 on fixed and permeabilized cells, enabling intracellular staining. Aggressive tissue dissociation may reduce surface CD163; intracellular detection is recommended in such cases.

Applications Reported: Flow cytometric analysis.
Applications Tested: Tested on mouse splenocytes at ≤0.25 µg/test (100 µL reaction volume). Cell number may range from 10⁵–10⁸ cells/test. Optimal titration is recommended.

Fluorochrome Information:
Super Bright 600 is a tandem dye excited at 405 nm and emits at 600 nm. Use a 610/20 bandpass filter and ensure instrument compatibility.

Staining Recommendations:
When using multiple Super Bright dye-conjugated antibodies, use Super Bright Complete Staining Buffer (Product # SB-4401) to minimize non-specific interactions.

Light Sensitivity: Protect vial and stained samples from light.
Fixation: Samples may be stored in IC Fixation Buffer (Product # 00-8222-49) or 1-step Fix/Lyse Solution (Product # 00-5333-57) for up to 3 days at 4°C in the dark.

Target Information

CD163 (M130 antigen, Ber-Mac3, Ki-M8, SM4) is a 130 kDa membrane glycoprotein, member of the scavenger receptor cysteine-rich superfamily, and receptor for hemoglobin-haptoglobin complexes. It protects tissues from oxidative damage and mediates uptake and recycling of iron.

CD163 is expressed on human monocytes and macrophages during late inflammation, upregulated by IL-10, IL-6, and dexamethasone, and shed upon LPS or PMA stimulation. It binds hemoglobin/haptoglobin complexes in a calcium- and pH-dependent manner, inducing intracellular signaling cascades and cytokine secretion (IL6, CSF1).
